P21

Also known as · P021 · Ciliary neurotrophic factor peptide mimetic

Overview

A small, blood-brain-barrier-penetrant peptide mimetic of ciliary neurotrophic factor, studied in rodent models of Alzheimer's disease for neurogenesis and reduced tau pathology.

Research profile · for clinicians

Based on research: entirely preclinical. Rodent work reports enhanced hippocampal neurogenesis, increased BDNF, and reduced tau hyperphosphorylation — with no human trials to date.

Mechanism of action

Mimics the active region of CNTF, increasing mature BDNF and TrkB signalling, promoting adult hippocampal neurogenesis, and inhibiting GSK-3β-driven tau phosphorylation.

Research record

  • [01]Improved cognition and reduced tau pathology in 3xTg-AD mice.
  • [02]No published human safety or efficacy data.
